Elle est en activité pendant et après la Prohibition. Le club a changé d'adresse à plusieurs reprises lors de son histoire.
Alors que le club se distingue par les meilleurs musiciens noirs de l'époque tels que Duke Ellington, Cab Calloway, Louis Armstrong ou Ethel Waters, il refuse généralement l'entrée aux Noirs. Durant l'âge d'or, le club est utilisé en tant que lieu de rendez-vous chic dans le cœur de Harlem, présentant régulièrement les dimanches les célébrités noctambules tel que Jimmy Durante, George Gershwin, Al Jolson, Mae West, Irving Berlin, Moss Hart, le maire de New York Jimmy Walker et bien d'autres encore.
Le champion poids lourd Jack Johnson ouvre le Club de Luxe à la 142e rue et l'Avenue Lenox à Harlem en 1920. Owney Madden (en), un contrebandier et gangster connu reprend le club en 1923 alors qu'il est emprisonné à Sing Sing et change son nom en Cotton Club. Le club est fermé brièvement en 1925 à cause de la vente d'alcool, mais rouvre plus tard sans ennuis. Les danseuses et stripteaseuses se produisent à la prison de Sing Sing où est emprisonné Owney Madden, jusqu'à sa libération en 1933.
