Este una dintre vechile lăcaşuri de cult din republică şi din Chişinău, care purta hramul Naşterea Maicii Domnului, anul edificării fiind de fapt incert. Tradiţia ştiinţifică numeşte anul 1752, iar păstrată în popor indică drept ctitor pe pârcălabul Chişinăului Vasile Mazarachi, serdar şi vistiernic, atestat în Chişinău în 1739 şi 1741. Este situată pe teritoriul oraşului Vechi, pe un promontoriu de pe malul râului Bâc, construită deasupra Fântânii Mari, cunoscut drept Fontalul oraşului, cu canotaţii deosebite în istoria aşezării. În prima fază, biserica, atestată în februarie 1789, avea hramul Sf. La sfârşitul secolului al XIX-lea primarul oraşului, K. Schmidt a înaintat iniţiativa construcţiei unui sediu nou pentru Duma orăşenească, pe locul ocupat de depoul de pompieri. Proiectarea a fost încredinţată arhitectului oraşului de atunci Mitrofan Elladi. În ajutorul său a fost invitat A. I. Bernardazzi. Arhitectura clădirii este în spirit eclectic în baza arhitecturii renaşterii italiene. Construcţia sediului Dumei a fost terminată în 1902. În timpul celui de al doilea război mondial, clădirea a fost deteriorată puternic, fiind reconstruită în anii postbelici după fotografiile clădirii, responsabil fiind arhitectul oraşului din acest timp R. Kurtz.
